- Counterion content. Peptides synthesized by solid-phase methods usually carry a residual counterion, most often acetate or trifluoroacetate. It should be identified and quantified.
- Appearance and solubility. Descriptive, but useful. If your receiving inspection sees something different from what the COA describes, that is a flag worth raising before the material goes into inventory.
- Test date and testing party. When the testing happened and who performed it. In-house versus independent third-party matters, and a COA should not make you guess.
How to Read a Peptide COA in About Four Minutes
1. Match the lot.
Compare the lot number on the COA to the lot printed on the physical packaging. If you are reviewing before purchase, ask which lot is currently shipping and confirm the COA you were sent corresponds to it.
2. Check the MS result against the theoretical mass.
The observed and theoretical molecular weights should line up. A mismatch is an identity problem, and identity problems are not something you resolve by email later.
3. Read the HPLC method, not just the percentage.
Column, gradient, detection wavelength. A purity figure with no method behind it is a claim, not a measurement.
4. Find the peptide content number.
If it is absent, ask for it. A supplier who cannot produce it for a given lot is telling you something about their manufacturing relationship.
5. Check the date.
A COA dated two years ago for a compound you are ordering this quarter means you are probably not looking at your lot.
6. File it against the order.
Store the COA with the purchase order and the receiving record. Documentation that exists but cannot be located during an audit does not help you.
Red Flags on a Peptide Certificate of Analysis
- No lot number, or a lot number that is obviously generic. Placeholder text like "representative lot" or "typical batch" defeats the purpose of the document.
- The same COA reused across multiple orders. If you order three times over six months and receive an identical PDF each time, either you are getting the same lot repeatedly or the supplier is not tracking lots at all.
- Scanned image with no source detail. A photocopied number with no lab letterhead, method, or analyst is not verifiable.
- Purity reported to an implausible precision. Reverse-phase HPLC area percent is not a measurement that meaningfully resolves to four decimal places.
- A COA that includes usage guidance. Analytical documentation describes the material. The moment a test report starts discussing administration, the supplier has drifted outside research-use framing and you have inherited some of that exposure.

Prioritize documentation you could hand to a medical board and defend line by line: which lot was used for which patient, when it was received, and what the COA said about it at the time.
